Em 1957, a DAF lançou a série 2000 DO. Este foi projetado para atender à crescente demanda em caminhões internacionais transfronteiriços, e foi inicialmente equipado com motores de 0,6 litros, construídos com Leyland, de 11,1 litros, mas posteriormente pelo motor DP 680 de 220 cv. O 2000 DO tinha um eixo traseiro de 10 toneladas projetado para operação com um peso bruto de 35 toneladas, o que era permitido na Alemanha e em muitos outros países europeus, exceto na Holanda.
Sua cabina era essencialmente uma versão modificada daquela introduzida nas séries 1300, 1600, T1800 e 1900., com uma grande grade na parte dianteira para auxiliar o resfriamento do motor. No entanto, devido a uma série de alterações na legislação europeia de camionagem, o DO 2000 não foi o grande sucesso que deveria ter sido.
Uma versão de cabina longa da DO 2000 foi introduzida em 1961, juntamente com um modelo AS 6x2 para transporte pesado. A gama de modelos foi expandida novamente em 1963 com a introdução da série 2300 DO. Ambas as gamas partilhavam o mesmo estilo de cabina e os motores Leyland 0.680, mas têm eixos diferentes, dando diferentes categorias de peso, com o 2300 DO avaliado até 35 toneladas.
